Indivumed performs for customers Protein Biomarker discovery studies based on a special proteomic platform and using Indivumed’s unique biobank samples and data base.
Two different approaches are available:
Profiling of intact proteins using fractionation techniques such as ionic exchange chromatography in combination with MALDI-TOF MS or by combination of HPLC (high performance liquid chromatography) and MALDI-TOF mass spectrometry (LC-MALDI-TOF MS). A combination of LCM (laser capture microdissection), HPLC and MALDI-TOF MS (LCM LC-MALDI) allows differentiated and comprehensive tissue studies.
Biomarker discovery by tryptic protein digestion (serum, tissue sections, LCM preparations), followed by liquid chromatography and MALDI-TOF MS measurement of tryptic fragments;
For purification and identification of potential biomarkers available technology includes ionic exchange chromatography and RP-HPLC followed by gel electrophoresis and enzymatic digestions of separated proteins. Results from peptide mass fingerprinting (performed by MALDI-MS) can be analyzed in house on a MASCOT server. For final identification, peptide fragments will be sequenced by MALDI-MS/MS;
For validation, different technologies, such as western blotting, ELISA or immunohistochemistry are available.
For statistical analysis approx. 300 clinical data points from each patient sample offer a unique opportunity to obtain clinical meaningful results by using Indivumed’s data base indivuNET and bioinformatic and statistical tools. Indivumed’s capability to perform within its clinical network validation studies within a short period of time provides another advantage for customers when collaborating with Indivumed for biomarker identification and validation.
